1. Introduction

Solubility is a fundamental property that affects every aspect of peptide handling and experimentation. This guide provides practical guidance for dissolving peptides in common solvents.

2. Solubility Fundamentals

Peptide solubility depends on: - Amino acid composition (hydrophilic vs. hydrophobic) - Net charge at the solvent pH - Counter-ion present (TFA vs. acetate) - Peptide concentration - Temperature

3. Water

Sterile water-for-injection is suitable for most hydrophilic and neutral peptides. Peptides rich in charged residues (Lys, Arg, Glu, Asp) typically dissolve readily in water.

4. BAC Water

Bacteriostatic water (0.9% benzyl alcohol) is suitable for multi-use vials. The preservative inhibits microbial growth. Not recommended for cell-based assays (benzyl alcohol may be cytotoxic).

5. Acetic Acid (0.1–1%)

Recommended for: - Hydrophobic peptides - Basic peptides (Arg, Lys-rich) - Peptides that form gels in water - Peptides with poor aqueous solubility

6. DMSO

Use for: - Concentrated stock solutions - Hydrophobic peptides - Peptides insoluble in aqueous solvents Important: Limit final DMSO to <0.1% in cell assays.

7. PBS (Phosphate-Buffered Saline)

Suitable for: - Most research applications - Cell culture studies - In vivo injections pH 7.4 matches physiological conditions.
